Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.59C>A (p.Thr20Asn), citing Ambry Variant Classification Scheme 2023: The p.T20N variant (also known as c.59C>A), located in coding exon 1 of the KIF1B gene, results from a C to A substitution at nucleotide position 59. The threonine at codon 20 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
