Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.2838C>A (p.Asp946Glu), citing Ambry Variant Classification Scheme 2023: The p.D900E variant (also known as c.2700C>A), located in coding exon 24 of the KIF1B gene, results from a C to A substitution at nucleotide position 2700. The aspartic acid at codon 900 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
