Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000218.3(KCNQ1):c.1871C>A (p.Thr624Asn), citing Ambry Variant Classification Scheme 2023: The p.T624N variant (also known as c.1871C>A), located in coding exon 16 of the KCNQ1 gene, results from a C to A substitution at nucleotide position 1871. The threonine at codon 624 is replaced by asparagine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
