NM_001378454.1(ALMS1):c.7309G>A (p.Glu2437Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2438K variant (also known as c.7312G>A), located in coding exon 8 of the ALMS1 gene, results from a G to A substitution at nucleotide position 7312. The glutamic acid at codon 2438 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
