Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_013372.7(GREM1):c.101C>T (p.Pro34Leu), citing Ambry Variant Classification Scheme 2023: The p.P34L variant (also known as c.101C>T), located in coding exon 1 of the GREM1 gene, results from a C to T substitution at nucleotide position 101. The proline at codon 34 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
