Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_012186.3(FOXE3):c.5C>G (p.Ala2Gly), citing Ambry Variant Classification Scheme 2023: The p.A2G variant (also known as c.5C>G), located in coding exon 1 of the FOXE3 gene, results from a C to G substitution at nucleotide position 5. The alanine at codon 2 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
