Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.5414C>T (p.Thr1805Ile), citing Ambry Variant Classification Scheme 2023: The p.T1797I variant (also known as c.5390C>T), located in coding exon 31 of the FLNA gene, results from a C to T substitution at nucleotide position 5390. The threonine at codon 1797 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
