Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000143.4(FH):c.1273G>T (p.Asp425Tyr), citing Ambry Variant Classification Scheme 2023: The p.D425Y variant (also known as c.1273G>T), located in coding exon 9 of the FH gene, results from a G to T substitution at nucleotide position 1273. The aspartic acid at codon 425 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
