This variant lies in the FH gene (transcript NM_000143.4) at coding-DNA position 915, deleting one base; at the protein level this means shifts the reading frame starting at phenylalanine residue 305, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.915delT pathogenic mutation, located in coding exon 7 of the FH gene, results from a deletion of one nucleotide at nucleotide position 915, causing a translational frameshift with a predicted alternate stop codon (p.F305Lfs*24). This variant has been observed in at least one individual with a personal and/or family history that is consistent with hereditary leiomyomatosis and renal cell cancer (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD).
