NM_001430.5(EPAS1):c.1694G>A (p.Ser565Asn) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S565N variant (also known as c.1694G>A), located in coding exon 12 of the EPAS1 gene, results from a G to A substitution at nucleotide position 1694. The serine at codon 565 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
