Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001430.5(EPAS1):c.1412G>A (p.Ser471Asn), citing Ambry Variant Classification Scheme 2023: The p.S471N variant (also known as c.1412G>A), located in coding exon 10 of the EPAS1 gene, results from a G to A substitution at nucleotide position 1412. The serine at codon 471 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
