NM_001430.5(EPAS1):c.1364C>T (p.Pro455Leu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P455L variant (also known as c.1364C>T), located in coding exon 10 of the EPAS1 gene, results from a C to T substitution at nucleotide position 1364. The proline at codon 455 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
