NM_177438.3(DICER1):c.4264GAG[6] (p.Glu1425_Ser1426insGluGlu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.4270_4275dupGAGGAG variant (also known as p.E1424_E1425dup), located in coding exon 22 of the DICER1 gene, results from an in-frame duplication of GAGGAG at nucleotide positions 4270 to 4275. This results in the duplication of 2 extra residues (EE) between codons 1424 and 1425. These amino acid positions are not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
