Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.1561T>C (p.Cys521Arg), citing Ambry Variant Classification Scheme 2023: The p.C521R variant (also known as c.1561T>C), located in coding exon 11 of the CTNNA3 gene, results from a T to C substitution at nucleotide position 1561. The cysteine at codon 521 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
