Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.2180_2181del (p.Thr727fs), citing Ambry Variant Classification Scheme 2023: The c.2180_2181delCA pathogenic mutation, located in coding exon 14 of the CTNNA1 gene, results from a deletion of two nucleotides at nucleotide positions 2180 to 2181, causing a translational frameshift with a predicted alternate stop codon (p.T727Rfs*6). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
