NM_000093.5(COL5A1):c.3514G>C (p.Asp1172His) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1172H variant (also known as c.3514G>C), located in coding exon 44 of the COL5A1 gene, results from a G to C substitution at nucleotide position 3514. The aspartic acid at codon 1172 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
