Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005502.4(ABCA1):c.8G>A (p.Cys3Tyr), citing Ambry Variant Classification Scheme 2023: The p.C3Y variant (also known as c.8G>A), located in coding exon 1 of the ABCA1 gene, results from a G to A substitution at nucleotide position 8. The cysteine at codon 3 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
