NM_000492.4(CFTR):c.649G>C (p.Glu217Gln) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E217Q variant (also known as c.649G>C), located in coding exon 6 of the CFTR gene, results from a G to C substitution at nucleotide position 649. The glutamic acid at codon 217 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
