Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.4335C>G (p.Asp1445Glu), citing Ambry Variant Classification Scheme 2023: The p.D1445E variant (also known as c.4335C>G), located in coding exon 27 of the CFTR gene, results from a C to G substitution at nucleotide position 4335. The aspartic acid at codon 1445 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
