This variant lies in the CDKN1B gene (transcript NM_004064.5) at coding-DNA position 429, deleting one base. Submitter rationale: The c.429delG pathogenic mutation, located in coding exon 1 of the CDKN1B gene, results from a deletion of one nucleotide at nucleotide position 429, causing a translational frameshift with a predicted alternate stop codon (p.L144*). This variant has been observed in at least one individual with a personal and/or family history that is consistent with CDKN1B-related multiple endocrine neoplasia (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
