NM_000719.7(CACNA1C):c.3200C>A (p.Ala1067Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A1067E variant (also known as c.3200C>A), located in coding exon 25 of the CACNA1C gene, results from a C to A substitution at nucleotide position 3200. The alanine at codon 1067 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
