Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.3124A>T (p.Thr1042Ser), citing Ambry Variant Classification Scheme 2023: The p.T1042S variant (also known as c.3124A>T), located in coding exon 23 of the BUB1B gene, results from an A to T substitution at nucleotide position 3124. The threonine at codon 1042 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
