Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.1612T>G (p.Tyr538Asp), citing Ambry Variant Classification Scheme 2023: The p.Y538D variant (also known as c.1612T>G), located in coding exon 14 of the BUB1 gene, results from a T to G substitution at nucleotide position 1612. The tyrosine at codon 538 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
