NM_001082486.2(ACD):c.435G>C (p.Gln145His) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q231H variant (also known as c.693G>C), located in coding exon 5 of the ACD gene, results from a G to C substitution at nucleotide position 693. The glutamine at codon 231 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
