Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.543A>C (p.Glu181Asp), citing Ambry Variant Classification Scheme 2023: The p.E181D variant (also known as c.543A>C), located in coding exon 6 of the BRCA1 gene, results from an A to C substitution at nucleotide position 543. The glutamic acid at codon 181 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
