This variant lies in the IDS gene (transcript NM_000202.8) at coding-DNA position 253, where G is replaced by T; at the protein level this means replaces alanine at residue 85 with serine — a missense variant. Submitter rationale: Variant summary: IDS c.253G>T (p.Ala85Ser) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 110674 control chromosomes (gnomAD). c.253G>T has been reported in the literature in individuals affected with Mucopolysaccharidosis Type II (Hunter Syndrome) (Froissart_1998, Froissart_2007, Semyachkina_2021). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.253G>A, p.Ala85Thr), supporting the critical relevance of codon 85 to IDS protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
