Pathogenic for Congenital heart defects, multiple types, 2 — the classification assigned by Victorian Clinical Genetics Services, Murdoch Childrens Research Institute to NM_001292034.3(TAB2):c.379C>T (p.Gln127Ter), citing ACMG Guidelines, 2015: Based on the classification scheme VCGS_Germline_v1.3.4, this variant is classified as Pathogenic. Following criteria are met: 0103 - Loss of function (LoF) and gain of function (GoF) are known mechanisms of disease in this gene. LoF is associated with congenital heart defects (CHD), nonsyndromic, 2 (MIM#614980) and congenital heart defects, multiple types, 2 (MONDO:0014000), while partial LoF appears to be associated with an emerging developmental delay and hypotonia syndrome with mild or no CHD (PMID: 36229919). GoF has an emerging association with frontometaphyseal dysplasia; however, this has currently only been observed in two individuals (PMID: 36229919). (I) 0107 - This gene is associated with autosomal dominant disease.
