This variant lies in the GFI1 gene (transcript NM_005263.5) at coding-DNA position 765, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 255 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Based on the classification scheme VCGS_Germline_v1.3.4, this variant is classified as VUS-3B. Following criteria are met: 0104 - Dominant negative is a known mechanism of disease in this gene and is associated with neutropenia, severe congenital 2, autosomal dominant (MIM#613107).
