Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.297C>A (p.Phe99Leu), citing Ambry Variant Classification Scheme 2023: The p.F99L variant (also known as c.297C>A), located in coding exon 1 of the FLNA gene, results from a C to A substitution at nucleotide position 297. The phenylalanine at codon 99 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
