Likely pathogenic for Ethylmalonic encephalopathy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_014297.5(ETHE1):c.263C>T (p.Ser88Leu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ETHE1 c.263C>T (p.Ser88Leu) results in a non-conservative amino acid change located in the Metallo-beta-lactamase (IPR001279) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249554 control chromosomes. c.263C>T has been reported in the literature as biallelic homozygous or compound heterozygous genotypes in individuals affected with Ethylmalonic Encephalopathy (example, Alfadhel_2016, Lim_2021). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as likely pathogenic.
