Submitter rationale: Variant summary: DMD c.11046+1G>A is located in a canonical splice-site within the last intron of the gene and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. However, it is not expected to undergo nonsense-mediated decay and to our knowledge, no pathogenic variants have been identified affecting the same donor site or within the last exon of the gene. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 182338 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.11046+1G>A in individuals affected with Dystrophinopathies and no experimental evidence demonstrating its impact on protein function have been reported.
