This variant lies in the FBN1 gene (transcript NM_000138.5) at 10 bases into the intron immediately before coding-DNA position 3713 through coding-DNA position 3713, replacing the reference sequence with AAAC. Submitter rationale: Variant summary: FBN1 c.3713-10_3713delinsAAAC is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 31394 control chromosomes.
