Uncertain significance for Primary dilated cardiomyopathy — the classification assigned by Petrovsky National Research Centre of Surgery, The Federal Agency for Scientific Organizations to NM_001018005.2(TPM1):c.88A>G (p.Lys30Glu), citing ACMG Guidelines, 2015: The NM_001018020.2:c.88A>G (p.Lys30Glu) variant in the TPM1 gene was found in the sporadic pediatric case (female) of DCM. Patient died at 5.5 y.o. due to progressive HF. This variant is absent in The Genome Aggregation Database (gnomAD) v2.1.1 and v4.1.0 (Date of access 05-07-2024). This variant has not been previously reported in any study to our knowledge. Most in silico predictors show pathogenic result of the protein change (varsome.com). In accordance with ACMG(2015) criteria variant is classified as Variant of Uncertain Significance (VUS) with following criteria selected: PM2, PM6, PP3.
