NM_001080453.3(INTS1):c.2984C>T (p.Ser995Leu) was classified as Uncertain significance for Neurodevelopmental disorder with cataracts, poor growth, and dysmorphic facies by Neuberg Centre For Genomic Medicine, NCGM, citing ACMG Guidelines, 2015: The observed missense variant c.2984C>T(p.Ser995Leu) in INTS1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is reported with the allele frequency 0.002% in the gnomAD Exomes. The amino acid Serine at position 995 is changed to a Leucine changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted as damaging by SIFT. The residue is conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
