Pathogenic for Ethylmalonic encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000019.9:g.(?_44015569)_(44015738_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 4 of the ETHE1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in ETHE1 are known to be pathogenic (PMID: 14732903, 19136963). A similar copy number variant has been observed in individuals with ethylmalonic encephalopathy (PMID: 14732903, 16183799, 20528888). For these reasons, this variant has been classified as Pathogenic.
