It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MCOLN1 are known to be pathogenic (PMID: 11030752, 11317355). For these reasons, this variant has been classified as Pathogenic. This variant is also known as del6433bp and g.511_6943del. A similar copy number variant has been observed in individuals with mucolipidosis type IV (PMID: 11030752, 21228398).
