Pathogenic for Aicardi-Goutieres syndrome 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000020.10:g.(?_35559143)_(35559298_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 5 of the SAMHD1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in SAMHD1 are known to be pathogenic (PMID: 19525956, 22461318). A similar copy number variant has been observed in individual(s) with clinical features of SAMHD1-related conditions (PMID: 33967934). For these reasons, this variant has been classified as Pathogenic.
