Pathogenic for Early-infantile DEE — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000020.10:g.(?_62027832)_(62045449_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 15-17 and part of exon 14 (c.1623_*10165del) of the KCNQ2 gene. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. This variant has not been reported in the literature in individuals with KCNQ2-related conditions. This variant disrupts the C-terminus of the KCNQ2 protein. Other variant(s) that disrupt this region (p.Cys744Leufs*91) have been determined to be pathogenic (PMID: 23692823). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic.
