Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000021.8:g.(?_43791362)_(43796709_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 12-13 and part of exon 11 (c.1135_*1509del) of the TMPRSS3 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with TMPRSS3-related conditions. This variant disrupts a region of the TMPRSS3 protein in which other variant(s) (p.Met448Thr) have been determined to be pathogenic (PMID: 28566687, 31412945, 32860223). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
