Likely pathogenic for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000021.8:g.(?_47532747)_(47533970_?)del, citing Invitae Variant Classification Sherloc (09022015): Loss-of-function variants in COL6A2 are known to be pathogenic (PMID: 19884007, 20976770). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant has not been reported in the literature in individuals with COL6A2-related disease. This variant is a deletion of the genomic region encompassing part of exon 5 (c.735+9_784del) of the COL6A2 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product.
