Pathogenic for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000021.8:g.(?_47552285)_(47554633_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the COL6A2 protein. Other variant(s) that disrupt this region (p.Tyr1002*) have been determined to be pathogenic (PMID: 29419890). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This variant has not been reported in the literature in individuals with COL6A2-related conditions. This sequence change is a complex rearrangement that is expected to disrupt part of exon 28 of the COL6A2 gene (c.2879_*1894). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 60 amino acids of the COL6A2 protein.
