This variant disrupts a region of the USH2A protein in which other variant(s) (p.Ser1136Asn) have been determined to be pathogenic (PMID: 22135276, 25991456, 27460420, 27957503, 28512305, 30718709). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with USH2A-related conditions. This variant results in the deletion of part of exon 17 (c.3317-1854_3449del) of the USH2A gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in USH2A are known to be pathogenic (PMID: 10729113, 10909849, 20507924, 25649381).
