Uncertain significance for Immunodeficiency 14 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_9779694)_(9781917_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 10-15 and part of exon 16 (c.1243-285_2054) of the PIK3CD gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in PIK3CD cause disease. This variant has not been reported in the literature in individuals affected with PIK3CD-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
