Uncertain significance for Catecholaminergic polymorphic ventricular tachycardia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_237674968)_(237675111_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 24 of the RYR2 gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in RYR2 cause disease. This variant has not been reported in the literature in individuals affected with RYR2-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
