Pathogenic for Medium-chain acyl-coenzyme A dehydrogenase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.10:g.(?_76226787)_(76228448_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 11-12 of the ACADM gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. A similar copy number variant has been observed in individual(s) with medium chain acyl-CoA dehydrogenase (PMID: 7633427). It has also been observed to segregate with disease in related individuals. This variant disrupts the p.Lys329, p.Ile416 amino acid residue in ACADM. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 15832312, 16617240, 16737882, 16763904, 20434380, 23028790, 23430840, 26947917). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
