Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.11:g.(?_227898106)_(227898217_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 38 of the COL4A4 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individual(s) with hematuria (Invitae). In at least one individual the variant was observed to be de novo. This variant disrupts a region of the COL4A4 protein in which other variant(s) (p.Gly1187Arg) have been observed in individuals with COL4A4-related conditions (PMID: 33532864, 33838161). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
