Pathogenic for Metachromatic leukodystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000022.10:g.(?_51062002)_(51064623_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exon(s) 6-8 and part of exon 5 (c.938_*1571del) of the ARSA gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with ARSA-related conditions. This variant disrupts a region of the ARSA protein in which other variant(s) (p.Thr410Hisfs*16) have been determined to be pathogenic (PMID: 7635478, 19606494). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
