NC_000003.11:g.(?_182788836)_(182792878_?)del was classified as Pathogenic for 3-methylcrotonyl-CoA carboxylase 1 deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the MCCC1 protein in which other variant(s) (p.Gly209Val) have been determined to be pathogenic (PMID: 25382614; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with MCCC1-related conditions. This variant results in the deletion of exons 5-6 and part of exon 7 (c.370-2603_712del) of the MCCC1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in MCCC1 are known to be pathogenic (PMID: 11181649, 15359379, 22642865).
