This copy number gain occurs in tandem (c.1925_3347dup) and is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in LAMB2 are known to be pathogenic (PMID: 15367484). This variant has not been reported in the literature in individuals affected with LAMB2-related conditions. A similar copy number gain has been confirmed to occur in-tandem in at least one individual (Invitae). This increases the likelihood that this variant disrupts protein function. For these reasons, this variant has been classified as Pathogenic.
