Loss-of-function variants in VHL are known to be pathogenic (PMID: 8956040, 12202531). This variant has not been reported in the literature in individuals with VHL-related conditions. This variant is a gross deletion of the genomic region encompassing part of exon 1 of the VHL gene (c.-3089_300del), which includes the initiator codon. This is expected to result in an absent or disrupted protein product.
